DETROIT A Wayne State University professor is leading research efforts to develop new treatment options to slow the progression of Parkinson's disease, a neurodegenerative disorder that affects nearly six million people around the world, with 50,000 to 60,000 new cases diagnosed in the United States alone each year. Symptoms of Parkinson's disease may include tremor, slowed motion, rigid muscles, difficulty initiating movement, speech changes, postural instability and more.
Currently no ideal therapies are available for slowing the degeneration process while relieving symptomatic abnormalities associated with Parkinson's disease. Aloke Dutta, Ph.D., professor of pharmaceutical sciences in WSU's Eugene Applebaum College of Pharmacy and Health Sciences, is hoping to change that with a recently awarded $2.15 million research grant, Novel Neuroprotective Treatment for Parkinson's Disease, from the National Institute of Neurological Disorders and Stroke of the National Institutes of Health. Dutta and his team aim to develop multifunctional therapeutic agents that will be useful not only in symptomatic treatment of Parkinson's disease, but also as disease-modifying agents promoting survival of the dopamine neurons that would otherwise gradually degenerate in Parkinson's disease.
According to Dutta, the etiology of Parkinson's disease is poorly understood, but both oxidative stress and mitochondrial dysfunction resulting from loss of glutathione coupled with increased concentration of free iron have been strongly implicated in dopamine cell death. In this regard, alpha-synuclein, a protein involved in fibrillization, has also been connected to Parkinson's disease.
"It is increasingly evident that for a complex disease such as Parkinson's, a drug targeting only one specific site will only partially address the therapeutic needs of the disease," said Dutta. "A growing body of literature suggests that suitable multifunctional drugs might be more effective to act as disease modifying agents. The overall goal of our research is to develop multifunctional therapeutic agents which will be useful not only in symptomatic treatment, but also as disease-modifying agents promoting survival of dopamine neurons."
"Our initial studies indicate facile blood-brain barrier crossing ability of such compounds developed by us," said Dutta. "The lead drugs we are developing have shown positive results in animal models of Parkinson's disease, and one of our lead molecules is exhibiting a neuroprotection property that may be critical to slowing the further progression of Parkinson's disease while being symptomatically beneficial in alleviating motor dysfunction."
The team aims to begin pre-clinical validation of these treatment agents soon. If results are positive, Dutta's study would shift the current research paradigm of Parkinson's disease by expanding treatment beyond that of acute motor defects stemming from dopamine deficiency.

For decades, ecologists have toiled to nail down principles explaining why some habitats have many more plant and animal species than others.
Much of this debate is focused on the idea that the number of species is determined by the productivity of the habitat.Shouldn't a patch of prairie contain a different number of species than an arid steppe or an alpine tundra?
Maybe not, says an international team of scientists that pooled its resources to re-evaluate the relationship between species numbers and habitat productivity.
The innovative, standardized global sampling of 48 sites on five continents yielded an unprecedented data set.
"Our study shows no clear relationship between productivity and the number of plant species in small study plots," says Utah State University plant ecologist Peter Adler.
Scientists say Adler's and his colleagues' findings represent a significant advance in ecological thought. The findings appear in this week's issue of the journal Science.
"We challenged a prevailing model developed in the early 1970s by British ecologist J. Philip Grime," says Adler, lead author of the paper. "He proposed that the number of species rises then declines with increasing productivity."Though hotly debated, this "hump-shaped" model has remained a textbook standard for nearly four decades.
"In the search for underlying principles of ecology in a very complex natural world, it's inevitable that even long-standing and accepted theories will be debunked as more data are accumulated and synthesized," says Henry Gholz, program director in the National Science Foundation's (NSF) Division of Environmental Biology, which funded the research.
In an "Emperor's New Clothes" moment, Adler remembers skeptical observations about the hump-shaped model made by graduate students in his classroom.
"'Why do ecologists spend so much time on this model when the evidence to support it is so weak?' they asked me," he says. "That was the kick I needed to pursue this question."
The challenge was daunting.
Existing, disparate case studies couldn't conclusively support Grime's unimodal pattern. Inconsistencies in data collection methods further hampered efforts to distill evidence to support the hump-shaped model. So Adler and fellow ecologists formed the Nutrient Network, or "NutNet," an NSF Research Coordination Network dedicated to investigating biodiversity and ecosystem processes in grasslands around the world.>Based at the University of Minnesota (UMN), the network is funded by an NSF grant to network organizers and UMN scientists Elizabeth Borer and Eric Seabloom.
"Our work not only sheds light on this classic question, it also demonstrates the power of a network approach," Borer says. "NutNet data are poised to inform many pressing ecological questions. Similar global, grassroots collaborations could help settle other longstanding scientific debates."
Says Gholz, "Research Coordination Networks are designed to facilitate these types of insights into the functioning of nature, insights that aren't possible in a focus on individual ecosystems."
Adler says that NutNet's data "emphasize the need to consider many factors to explain patterns of diversity--not just productivity alone."
NutNet's findings should spur ecologists to focus on other important factors regulating biodiversity, he says, such as evolutionary history, disturbance and resource supply.
"It's time to remove outdated models from our textbooks and concentrate on more sophisticated approaches," Adler says,"That will improve our ability to predict the effects of environmental change on biodiversity."

The grid's shortcomings have been well-documented, but efforts to modernize it haven't kept up with demand. Many electrical transmission lines are outdated, and parts of the grid date back to the time of Thomas Edison.
The chairman of the Federal Energy Regulatory Commission, which oversees the nation's grid, acknowledged increasing problems with the system.
In a July interview with ProPublica, FERC Chairman Jon Wellinghoff said that while the electric grid is reliable, it is degrading. "It's not getting better," he said. "It's getting worse."
Many experts say smart-grid technology would help. Such a system would be able to intelligently respond to sudden peaks or drops in demand and energy supply.
Last week, for example, a mishap involving a single worker doing repairs on a power station near Yuma, Ariz., led to rolling blackouts over parts of Arizona, Southern California and Northern Mexico. The short circuit caused San Diego County's power-supply system to completely shut down after it was required to take on the demand of those affected in Arizona and buckled under the extra load.

Wellinghoff, FERC's current chairman, said the commission also needs broader authority to oversee transmission lines. That would require congressional approval.
One project that might benefit from broader FERC power is the Susquehanna-Roseland power line. The line, which would run from Pennsylvania to Northern New Jersey, has been delayed for two years because a four-mile stretch passes through a national park. Although the line would follow the same path as a pre-existing line, the National Park Service has blocked construction to analyze the effect of the line on the environment. Wellinghoff said FERC's engineers had already done an environmental review.

Wholesale prices have doubled in some places as farmers nurse their surviving pumpkin plants toward a late harvest. Some farmers are trying to buy pumpkins from other regions to cover orders.
"I think there's going to be an extreme shortage of pumpkins this year," said Darcy Pray, owner of Pray's Family Farms in Keeseville, in upstate New York. "I've tried buying from people down in the Pennsylvania area, I've tried locally here and I've tried reaching across the border to some farmers over in the Quebec area. There's just none around."
Hurricane Irene raked the Northeast in late August, bringing torrents of rain that overflowed rivers and flooded fields along the East Coast and into southern Canada. Pray saw his entire crop, about 15,000 to 20,000 pumpkins, washed into Lake Champlain.
But pumpkin farmers had been having a difficult year even before the storm. Heavy rains this spring meant many farms had to postpone planting for two or three weeks, setting back the fall harvest, said Jim Murray, owner of the Applejacks Orchard in Peru, N.Y.
A late harvest can be fatal to business because pumpkin sales plummet after Halloween on Oct. 31. Wholesalers need to get pumpkins on their way to stores by mid-September.
Another spate of rain about two weeks before Irene caused outbreaks of the phytophthora fungus ?a type of water mold ? in many fields, said Jim Stakey, owner of Stakey's Pumpkin Farm in Aquebogue, on New York's Long Island.
This week a cold snap threatened to kill the surviving vines, Murray said.
"We were real close to a frost last night," Murray said Saturday. "It was 34, and if we had had a frost, a lot of immature pumpkins would have never made it."
The wholesale price for a bin of 32 to 45 pumpkins ranged from $150 to $200 in upstate New York, about twice the normal price, Pray said. It was still unclear how the shortage would affect retail prices, he said, but in a normal year, each pumpkin could sell for up to $15 at a supermarket in a big city like New York.
The problems for Northeast farmers have been a boon for growers in other parts of the country, especially in big pumpkin-producing states like Illinois, Indiana, California, Ohio and Michigan.

The pumpkin crunch could also affect tourism because pick-your-own pumpkin farms have become important attractions in many rural areas, farmers said.
Stakey's 26-acre farm offers pumpkin-picking along with pony rides, a cornfield maze, rides in a farm wagon and other events. He said he's buying extra pumpkins to put in the fields to supplement his own crop.
"Just get your pumpkins early, that's all I can say," he said. "It's going to be a difficult season."
